This fluorescent labeled antibody was purified from whole serum via Protein A affinity columns, and dialysis against PBS (137 mM NCI, 2.7 mM KCI, 4.3 mM Na2HPO4, 1.4 mM KH2PO4, pH 7.5). The HPLC purified standard peptide, corticotropin releasing factor (human, rat) was detected by this rhodamine labeled IgG (1: 20-40 i.e. 25-50 µl/ml) using FLx800 Microplate Fluorescence Reader (BIO-TEK INSTRUMENTS, INC) at wavelengths of 560/20 nm (excitation) and 620/40 nm (emission). We recommend that each laboratory determines an optimum titer for use in their particular assay system.
